Abstract

In the interest of more thoroughly understanding the relationship between sample preparation technique and spectra quality produced in Matrix-Assisted Laser Desorption/Ionization (MALDI) mass spectrometry, we have investigated details of the Electrospray (ES) process of sample preparation by observing sample morphology with Scanning Electron Microscopy (SEM), Atomic Force Microscopy (AFM) and Matrix-Enhanced Secondary Ion Mass Spectrometry (MESIMS). Electrospray deposition reduces the analyte segregation that can occur during tradional dry drop depostion for MALDI. Our investigations demonstrate improvements in the precision of MALDI average molecular mass measurements attributed to use of the ES sample preparation technique.
